DefinitionFKS1 encodes a catalytic subunit of the beta-1,3-D-glucan synthase complex typically associated with resistance to echinocandin antifungals. Although azoles primarily target ergosterol biosynthesis, rare FKS1 mutations have been reported to confer resistance. These mutations are thought to alter cell wall integrity and membrane to cell wall interactions, triggering compensatory stress response pathways that indirectly mitigate azole-mediated growth inhibition. Such cases represent uncommon, context-dependent resistance mechanisms rather than direct target-site resistance.
